個人簡介
陳玲玲,女,2009年畢業於美國康乃狄克大學醫學院獲生物醫學博士學位,同時獲商學院工商管理學碩士學位。同年5月作為獨立PI獲得 Connecticut Stem Cell Seed Award研究經費資助,受聘於康乃狄克大學幹細胞學院從事博士後研究,並於2010年5月任助理教授。2011年1月任上海生化與細胞所研究員。目前擔任國際期刊 Trends in genetics、 Genome biology、 Journal of Biological Chemistry編委。
2011年1月至今任中國科學院上海生命科學研究院生物化學與細胞生物學研究所研究員。主要從事RNA編輯和長非編碼RNA對基因表達調控和幹細胞命運決定的分子機制研究。
研究工作
人類基因組測序計畫發現多於98%的轉錄基因組序列為非編碼RNA(ncRNA)。這些ncRNA包括housekeepingnc RNA (如tRNAs,rRNAs和snRNAs等)、小RNA(如miRNAs和piRNAs等),以及上萬條長非編碼RNA(lncRNA,>200nt)。最近幾年來的研究表明lncRNAs廣泛參與一系列細胞的重要功能調控,包括細胞核亞結構的形成、基因表達的遺傳與表觀遺傳調控等。
實驗室主要研究哺乳動物細胞中lncRNAs的功能調控,包括新類型lncRNA分子的發掘、其加工成熟機制、以及它們在轉錄、染色體、細胞核亞結構和幹細胞乾性維持等方面的調控功能研究。未來五年,將主要研究以下內容:
1、通過轉錄組測序和計算生物學手段結合的方法,發掘和鑑定人類基因組中的新類型lncRNA分子;
2、利用分子生物學、細胞生物學和生物化學的手段,研究新lncRNA分子在基因表達調控中的作用機制;
3、用基因組編輯和活細胞成像等技術手段,研究lncRNA在細胞核亞結構和染色體維持和調控過程中功能;
4、以hESCs的多功能潛能性維持和分化為模型,研究lncRNA對人源胚胎幹細胞的命運決定的調控。
研究成果將能夠豐富對lncRNAs功能和機制的認識,也為hESCs的多功能性維持和命運改變提供一個全新水平的調控。
